Mechanism of Action

KPV inhibits NF-κB activation by directly entering cells and interacting with the p65 subunit of the NF-κB transcription factor, preventing nuclear translocation. This suppresses transcription of pro-inflammatory cytokines (IL-1β, TNF-α, IL-6), chemokines, and adhesion molecules. KPV also inhibits inflammasome activation and reduces IL-8 production in intestinal epithelial cells. Notably, this mechanism does not require melanocortin receptor binding, distinguishing KPV from full-length α-MSH and other melanocortin peptides.
