Mechanism of Action

MOTS-c activates AMPK (AMP-activated protein kinase) by inhibiting the folate cycle and de novo purine synthesis, leading to AICAR accumulation. Nuclear translocation of MOTS-c enables direct interaction with transcription factors governing antioxidant response elements (ARE) and stress-response genes. In skeletal muscle, MOTS-c promotes insulin-independent GLUT4 translocation and upregulates oxidative phosphorylation complexes.
