Orforglipron is a once-daily, non-peptide small-molecule agonist of the glucagon-like peptide-1 (GLP-1) receptor. It activates the receptor through a small-molecule binding mode distinct from peptide agonists, and unlike oral semaglutide it does not require a permeation/absorption enhancer or fasting and water restrictions. It is a GLP-1-only agent, distinct from tirzepatide, which is an injectable dual GIP/GLP-1 peptide.
