N-Acetyl Semax Amidate is a doubly end-modified analog of Semax — the heptapeptide (Met-Glu-His-Phe-Pro-Gly-Pro) built on the ACTH(4-7) fragment with a Pro-Gly-Pro tail in place of residues 8-10 — bearing N-terminal acetylation and C-terminal amidation. These terminal modifications are intended to increase resistance to aminopeptidase and carboxypeptidase degradation relative to unmodified Semax while preserving the parent peptide's neuromodulatory profile. Mechanism of Action
As a Semax analog, it is studied for the parent peptide's mechanisms: upregulation of brain-derived neurotrophic factor (BDNF), nerve growth factor (NGF), and TrkB receptor expression in hippocampal and cortical tissue, and modulation of the melanocortin system via MC4R. The N-terminal acetyl and C-terminal amide groups cap the peptide termini, a common strategy to slow exopeptidase cleavage; comparative stability versus unmodified Semax is itself a research endpoint.
