DSIP is a synthetic nonapeptide, Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, first isolated and sequenced in 1978.

The name records the context in which the peptide was originally isolated and does not describe a verified property of the material. The peptide was isolated from the extracorporeal dialysate of cerebral venous blood in rabbits and named at that time. A molecular weight of 849 is reported in the source literature, consistent with the PubChem record.

Structural characteristics

The original report describes synthesis of the nonapeptide together with five possible metabolic fragments, two nonapeptide analogues with one and two amino acid exchanges, and a related tripeptide. It records that the aspartyl residue at position 5 must be in the alpha-aspartyl form: the report states that only the pure alpha-aspartyl peptide showed the described activity, in contrast to its beta-Asp isomer.

Analytical considerations

The alpha-aspartyl versus beta-aspartyl distinction at position 5 is an identity consideration, not merely a synthesis detail: the two isomers share a molecular formula and mass, so mass spectrometry alone does not distinguish them and a sequence- or separation-based method is required. The single tryptophan residue provides a UV chromophore near 280 nm.
